二乙烯亚胺对脊髓灰质炎病毒Ⅱ型Sabin株的灭活效果Inactivation Effect of Binary Ethylenimine on Type Ⅱ Poliovirus Sabin Strain

摘要(Abstract):
目的观察二乙烯亚胺(Binary ethylenimine,BEI)对脊髓灰质炎病毒Ⅱ型Sabin株的灭活效果。方法在不同温度(37、33、28和25℃)下采用4种浓度(0.002、0.001、0.0005和0.00025mol/L)的BEI及0.0925mg/ml的甲醛对脊髓灰质炎病毒Ⅱ型Sabin株进行灭活,采用微量滴定法测定病毒感染性滴度,ELISA法检测灭活病毒液D抗原含量,并进行病毒灭活验证试验。结果在37℃条件下,0.002mol/L的BEI能在48h内完全灭活病毒,灭活后的病毒液D抗原含量回收率为90.6%,显著高于甲醛灭活的回收率(60.6%)。经验证,灭活彻底,无活病毒存在。结论 BEI对脊髓灰质炎病毒Ⅱ型Sabin株的D抗原破坏小,能较好地保持疫苗的有效成分。
