麻疹病毒属病毒种特异性探针的设计Design of Probes for Species Specificity of Viruses Belonging to Morbillivirus

摘要(Abstract):
目的设计能检测麻疹病毒、犬瘟热病毒、牛瘟病毒、海豹瘟热病毒、海豚瘟热病毒和小反刍兽疫病毒的基因芯片探针,制备芯片用于以上病毒的种特异性检测。方法在ncbi网站下载上述病毒全部基因组序列信息,利用生物学软件CLUSTAL W对从GenBank中获得的6种病毒所有序列进行比对,获得每种病毒有意义的特异序列,以此作为设计oligo探针的备选序列,利用生物软件Primer Premier 5.0设计探针。选择最优化探针,制备基因芯片,并进行特异性验证。结果获得6条长度均一,退火温度相近,特异性良好的种特异性寡核苷酸探针。结论利用GenBank数据库和生物学软件Primer Premier 5.0设计病毒检测基因芯片探针,是一种快速、有效的方法。
